aku kana kakai oleh tikus kelmarin... hahaha.. baruth ku pernah tu.. i think pasal they havent been handled before.. sal nya durang histology rats.. and i decided to do blood pressures on them.. kana kakai th plang.. hahaha.. did more expts again today.. finally abis 3 groups of my rats for langendorffs and organ baths.. *lap paluh* hahaha.. ngalih bah bnyk kraja.. alum lgi buat histolgy, blood and plasma analysis.. aduhh~~ kepisan... kepisan.. walaupun ku nada exam... the amount of work that i did per day can be considered as studying..just imagine.. lab work non stop for about 5hours.. no food.. no sitting down.. continuous hands on ... concentrate on experiments.. make sure nothing goes wrong.. as follows .. make sure....:

• get rats from animals house.. do body weight, water and food consumption first
• calibration for langendorff, blood pressure and organ baths are right
• do blood pressure on them first
  
• calibration for langendorff- no bubbles, make sure the value is not drifting.. if not .. calibrate again!
• calibration for blood pressure - make sure its the right pressure.. if not .. calibrate again!
• calibration for organ bath - make sure it has the right tension.. if not.. calibrate again!
• gases on to supply carbogen.. if not on.. expt wont work!
• prepare buffer.. if wrong concentration or chemical being put in.. expt wont work!
  
• get everything ready.. scissors.. scalpels.. forceps.. make sure everything is sharp.. if not u'll go slow!
• all water baths on! if not.. expts wont work!
• put tyrode solutions in organ baths and buffers for langendorff.. if not.. expt wont work!
• setup the channels for organ baths .. give labellings.. if not.. ure channels will be a mess and doesnt make any sense
• all syringes ready for lethabarb.. heparin.. blood.. if not.. it will slow u down..

atu baru before experiments even started.. now when the experiments starts... make sure...

• wear ure lab coat.. if not.. get blood stain all over ure fav shirt!
• get heparin in the femoral vein (payah .. really small) .. if not .. blood clot
• suck blood properly from the vena cava.. if not.. u wont get a good plasma after centrifuged!
• cut heart properly.. if not ... u end up with short aorta.. and expt wont go really well!
• thoracic aorta not stretched for organ baths.. if not.. inconsistent results
• the buffer is always filled up.. if not.. pressure not consistent..
• aorta is properly tied.. if not.. heart doesnt get sufficient buffer or buffer keep shooting out from the aorta.. and u get wet!
  
• put in balloon properly.. if not .. pops out.. do again!
  
• pace the heart at the right atrium.. if not.. doesnt give good results..
• record when u doing experiments.. if not useless!
• type in the numbers when recording.. if not useless again!

...... haha atu langendorff tu.. now... organ baths

• wash the vessels every 10mins.. in not.. nda ku brapa tau plang.. but supervisor slalu marah if nda buat ani.. slalu ku kana hahaha
• put 25ml of buffer in each... if not.. affects the concentration being put in..
• tension before about 10mN.. if not.. hard to compare..
• drugs out from the freezer awal.. if not.. baku masih!.. and alum dapat start expt.. so have to wash the vessels for every 10mins more..
• the pippette says the right volume.. if not.. expt hancur!.. do again!
  
• u put the right concentration of drug.. if not .. expt hancur.. do again!
• dont touch or tepigang cikit the thing holding the baths.. if not.. tension affected.. results useless!..
  
• type the right conc on the screen .. if not.. nanti hard to analyse
• when ure contracting.. makes ure ya stabilise before adding the next conc .. if not.. results bida!
• basically concentrate lah... yatah mambari paning tu.. hahaha

now organ weights...

• dont rip any of the organs.. if not .. not accurate
• get fats out of the organs... if not .. not accurate
• blot them dry.. if not.. not accurate
• for fats measurements.. make sure at the right places.. if not.. not accurate..
  

hahaha.. atu abis experiments.. now after semua abis

• get plasma out from centrifuged blood and put in the freezer .. if not.. no samples..
• clean off buffer.. and put in distilled water.. if not.. affect next expt
• balloon out ... if not it will dry out .. and make new one!
• cover transducer..
  
• save all files.. any paling important.. if not.. no results!
• wash all organ baths with distilled water.. if not.. affect next expt
• take out all vessels from wire.. if not.. next expt bnyk kraja lgi
• put drugs back in the freezer.. if not.. drugs can be used anymore..
  
• put buffers back in the fridge.. if not.. next time buat new one.. ngalih lagi~~
• gas off.. if not.. ntah.. waste of gas..
• switch off everything.. waste electricity..
• wash everything.. trays.. scissors.. scapels.. forceps..
  
• tutup lampung lab
• bring out the data for organ weights

yg ku cakap ani .. baru sal experiment day for langendorffs and organ baths.. alum lgi yg lain .. OGTT.. blood pressure.. histology, blood and plasma analysis... ppl might think.. apa jua ah... research.. nada exam.. nda payah blajar.. nda payah revision.. nda payah dtg lectures.. well I have to .. read journals.. write thesis.. attend seminars.. stats exam.. lab meetings.. do presentations.. do graphs.. analyse results.. what else ah... hahahah.. oh well.. yatah kehidupan Honours nie... nada kraja bah aku ani.. saja kn becerita hidup ku.. hahaha yatah namanya blogging nie... hahahah .. bah atu sja...
